DAG2plus, diepe multi-omics fenotypering van Lifelines Deep deelnemers – deel 3

With the advent of different single-cell multi-omics techniques, it is now possible to probe the human body at an unprecedented level. We have recently started generating single-cell RNA-seq data, and now want to generate additional (layers of) single-cell data to further increase the tremendous value that the multi-omics data in Lifelines Deep represents. We would like to collect blood samples from 200 Lifelines Deep individuals. Peripheral blood mononuclear cells (PBMCs) will be isolated from the blood and will be used to generate single-cell gene expression and gene regulatory information (i.e. scRNA-seq and scATAC-seq data). These data layers will be coupled to an individual’s genetic background (data is already available at the computer cluster of the Genetics department), which will enable us to obtain cell type-specific information about how genetics is affecting gene expression levels and gene regulation. By reconstructing personalized gene regulatory networks, we expect this information can aid in realizing precision medicine in the future.
